Draq5是一種遠(yuǎn)紅外熒光活細(xì)胞DNA染料,是一種對(duì)雙鏈DNA具有高親和力的蒽醌染料。它是一種可以透過(guò)細(xì)胞膜的染料,可標(biāo)記活細(xì)胞或固定后/死細(xì)胞。在流式細(xì)胞術(shù)中,這種染料可用于區(qū)分有核和無(wú)核細(xì)胞。由于Draq5能夠按照化學(xué)計(jì)量比結(jié)合至DNA,因此還可用于報(bào)告細(xì)胞核DNA含量,適用于染色體倍數(shù)和細(xì)胞周期分析。在熒光顯微鏡分析中,它可用作細(xì)胞核復(fù)染劑。Draq5有很多應(yīng)用,高度兼容現(xiàn)有儀器平臺(tái)廣泛使用的程序,主要的應(yīng)用領(lǐng)域?yàn)镠CS,細(xì)胞模型,GFP,流式細(xì)胞儀和熒光顯微鏡。
Draq5的激發(fā)波長(zhǎng)范圍為488至647 nm。對(duì)于成像顯微術(shù),建議使用633或647 nm的光源進(jìn)行激發(fā)。對(duì)于流式細(xì)胞術(shù),在488 nm處激發(fā)這種染料時(shí),可使用685 LP二向分色鏡和710/50通道進(jìn)行檢測(cè);在633 nm處激發(fā)時(shí),可以使用660/20通道進(jìn)行檢測(cè)。對(duì)于細(xì)胞周期/DNA分析應(yīng)用,建議使用波長(zhǎng)較長(zhǎng)的濾光片,例如735 LP二向分色鏡和780/60通道來(lái)優(yōu)化G1和G2/M峰的CV值。請(qǐng)確保您的儀器能夠檢測(cè)該染料。
由于它的發(fā)射和激發(fā)波長(zhǎng)范圍很寬,不建議將Draq5與其他可被488或633 nm激光激發(fā)的遠(yuǎn)紅光熒光染料聯(lián)用。
產(chǎn)品濃度為5 mM,以5×104 cells/well為例,染色工作液濃度20 μM計(jì)算,200 μL的規(guī)格可以做400個(gè)樣本。
注意事項(xiàng)
1. 使用前請(qǐng)將產(chǎn)品瞬時(shí)離心至管底,再進(jìn)行后續(xù)實(shí)驗(yàn)。
2. 熒光染料均存在淬滅問題,請(qǐng)盡量注意避光,以減緩熒光淬滅。
